The Mononuclear Phagocyte System in Infectious Disease

The Mononuclear Phagocyte System in Infectious Disease

Autor: Geanncarlo Lugo-villarino , Céline Cougoule , Etienne Meunier , Yoann Rombouts , Christel Vérollet , Luciana Balboa

Número de Páginas: 790

The Mononuclear Phagocyte System (MPS) of vertebrates is composed of monocytes, macrophages and dendritic cells. Together, they form part of the first line of immune defense against a variety of pathogens (bacteria, fungi, parasites and viruses), and thus play an important role in maintaining organism homeostasis. The mode of transmission, type of replication and mechanism of disease-causing differ significantly for each pathogen, eliciting a unique immune response in the host. Within this context, the MPS acts as both the sentinel and tailor of the immune system. As sentinels, MPS cells are found in blood and within tissues throughout the body to patrol against pathogenic insult. The strategy to detect 'microbial non-self' relies on MPS to recognize conserved microbial products known as 'pathogen-associated molecular pattern' (PAMPs). PAMPs recognition represents a checkpoint in the response to pathogens and relies on conserved 'pattern recognition receptors' (PRRs). Upon PRR engagement, MPS mount a cell-autonomous attack that includes the internalization and compartmentalization of intracellular pathogens into toxic compartments that promote destruction. In parallel, MPS cells...

Metabolism of Drugs and Other Xenobiotics

Autor: Pavel Anzenbacher , Ulrich M. Zanger

Número de Páginas: 755

A practice-oriented desktop reference for medical professionals, toxicologists and pharmaceutical researchers, this handbook provides systematic coverage of the metabolic pathways of all major classes of xenobiotics in the human body. The first part comprehensively reviews the main enzyme systems involved in biotransformation and how they are orchestrated in the body, while parts two to four cover the three main classes of xenobiotics: drugs, natural products, environmental pollutants. The part on drugs includes more than 300 substances from five major therapeutic groups (central nervous system, cardiovascular system, cancer, infection, and pain) as well as most drugs of abuse including nicotine, alcohol and "designer" drugs. Selected, well-documented case studies from the most important xenobiotics classes illustrate general principles of metabolism, making this equally useful for teaching courses on pharmacology, drug metabolism or molecular toxicology. Of particular interest, and unique to this volume is the inclusion of a wide range of additional xenobiotic compounds, including food supplements, herbal preparations, and agrochemicals.

Drug Hypersensitivity

Drug Hypersensitivity

Autor: W. J. Pichler

Número de Páginas: 441

Drug hypersensitivities are immune-mediated reactions characterized by exanthems, fever and internal organ involvement. They represent a challenging field of research: many different drugs can induce a variety of clinical symptoms through multiple mechanisms of drug-host interaction, many of which are poorly understood. This is the first book to approach the phenomenon of drug hypersensitivity in a comprehensive manner. Besides epidemiological aspects, it addresses the immunological mechanisms underlying these complicated reactions which go far beyond the IgE-mediated drug allergies also considered in the book. Furthermore, the book covers clinical manifestations and new diagnostic methods, and introduces some recently established animal models. Well-accepted and also completely new concepts are presented and discussed in detail. Many topics are treated from multiple perspectives, and the 33 chapters are thoroughly cross-referenced. This publication will be of immense value not only to allergologists, dermatologists and anyone prescribing medication, but also to scientists in a pharmaceutical industry challenged by the economic effects of failures in drug development or drug...

Models of Protection Against HIV/SIV

Models of Protection Against HIV/SIV

Autor: Gianfranco Pancino , Guido Silvestri , Keith Fowke

Número de Páginas: 361

A successful vaccine for the prevention and/or immunotherapy against HIV/AIDS is one of the prominent challenges of the 21st century. To date, all human vaccine trials against this virus/disease have resulted in failure, or at best have shown very low efficacy. The scientific community dealing with HIV/AIDS has unanimously proposed a focus on basic science, with the intention of identifying correlates of protection that can serve as guides in developing and evaluating vaccine preparation. However, Nature seems to have already found several ways of dealing with infections by HIV and related primate lentiviruses, either by resisting infection or, once infected, avoiding immune damage and immunodeficiency. Models of Protection Against HIV/SIV will allow for an in-depth reflection on the perspectives for vaccine and therapy research derived from important recent studies. It will be authored by some of the most well known specialists in the field of HIV resistance/protection: including F. Barré-Sinoussi (2008 Nobel Prize for Medicine winner), B. Walker, S. Rowland-Jones, A. Telenti, M. Lederman and F. Plummer. This book is structured in a unique way, looking at three models of...

Natural Hosts of SIV

Natural Hosts of SIV

Autor: Aftab A. Ansari , Guido Silvestri

Número de Páginas: 415

Natural Hosts of SIV: Implications in AIDS thoroughly reviews the possible mechanisms by which African nonhuman primate natural hosts of lentiviruses remain essentially disease-free while other hosts exhibit disease and death. The book ultimately indicates directions for further research and potential translations of this compelling phenomenon into novel approaches to treat and prevent HIV. When Asian non-human primate non-natural hosts are experimentally infected with viruses isolated from African species, disease and death normally results. Meanwhile, these African nonhuman primate natural hosts maintain similar levels of plasma and cellular viremia and exhibit compellingly different, essentially disease-free, states. This work attempts to answer the question of how the natural host remains disease resistant. - Summarizes the past 30 years of research in this field and describes the latest developments in AIDS research using nonhuman primate animal models - Provides insights into how this large body of scientific work can be translated into novel approaches to treat and prevent HIV - Highlights the areas that merit future pursuit, focusing on potential applications for the...

New Research on Pharmacogenetics

New Research on Pharmacogenetics

Autor: Linda P. Barnes

Número de Páginas: 636

The terms pharmacogenomics and pharmacogenetics tend to be used interchangeably, and a precise, consensus definition of either remains elusive. Pharmacogenetics is generally regarded as the study of genetic variation that gives rise to differing response to drugs, while pharmacogenomics is the broader application of genomic technologies to new drug discovery and further characterization of older drugs. Pharmacogenetics considers one or at most a few genes of interest, while pharmacogenomics considers the entire genome. Much of current clinical interest is at the level of pharmacogenetics, involving variation in genes involved in drug metabolism with a particular emphasis on improving drug safety. This new book presents leading-edge research in this dynamic field.

Cien años después

Cien años después

Autor: Camacho Zarco, Aldo

Human Diversity

Human Diversity

Autor: Charles Murray

Número de Páginas: 721

All people are equal but, as Human Diversity explores, all groups of people are not the same -- a fascinating investigation of the genetics and neuroscience of human differences. The thesis of Human Diversity is that advances in genetics and neuroscience are overthrowing an intellectual orthodoxy that has ruled the social sciences for decades. The core of the orthodoxy consists of three dogmas: - Gender is a social construct. - Race is a social construct. - Class is a function of privilege. The problem is that all three dogmas are half-truths. They have stifled progress in understanding the rich texture that biology adds to our understanding of the social, political, and economic worlds we live in. It is not a story to be feared. "There are no monsters in the closet," Murray writes, "no dread doors we must fear opening." But it is a story that needs telling. Human Diversity does so without sensationalism, drawing on the most authoritative scientific findings, celebrating both our many differences and our common humanity.
